Target-mediated drug disposition

Nonlinear clearance for a drug may be due to these two mechanisms:

  • Receptor-mediated endocytosis which removes drug-receptor complex

  • Drug binds soluble targets to comprise a complex which is degraded via macrophage engulfment

These may be accurately described by the target-mediated drug disposition (TMDD) model. The default values in the simulator describe Ascrinvacumab PK in cynomolgus monkeys.
